Bilayer tablets represent a sophisticated oral drug delivery system that enables the incorporation of two different release profiles typically immediate release (IR) and sustained or extended release (SR/ER) within a single dosage form. This dual-release system is particularly advantageous for improving therapeutic efficacy, enhancing patient compliance, and reducing dosing frequency. The immediate release layer provides a rapid onset of action, delivering the drug quickly into the bloodstream to initiate therapeutic effects, while the sustained release layer maintains drug levels within the therapeutic window for an extended period, reducing the need for multiple daily doses. Additionally, bilayer tablets can be formulated to combine two different active pharmaceutical ingredients (APIs) with distinct pharmacokinetic profiles, enabling combination therapy in a single tablet. These systems are especially beneficial in the management of chronic conditions such as hypertension, diabetes, and pain, where controlled drug release is essential for optimal treatment outcomes.
The formulation and manufacturing ”f bilayer tablets require precise control over layer uniformity, adhesion between layers, and drug release kinetics, often involving specialized equipment and polymers to achieve the desired performance. Despite the challenges, bilayer tablet technology continues to evolve, offering a promising platform for the development of advanced oral drug delivery systems that align with the growing demand for more effective and patient-friendly medications.
