Abstract :

Breast cancer is one of the most prevalent malignancies worldwide and remains a leading cause of mortality among women. The Epidermal Growth Factor Receptor (EGFR) has emerged as a critical molecular target due to its overexpression in aggressive forms of breast cancer, particularly triple-negative breast cancer. Pyrimidine-based heterocyclic compounds have gained significant attention in medicinal chemistry owing to their structural versatility and biological relevance. This review provides a comprehensive discussion on the design, synthesis, and biological evaluation of pyrimidine derivatives as EGFR inhibitors. It also highlights structure–activity relationships, mechanisms of action, and future prospects in the development of effective anticancer therapies.
