REVIEW ON MICROSPONGE-BASED TOPICAL DELIVERY OF NSAIDS

Abstract :
Non-steroidal anti-inflammatory drugs (NSAIDs) are widely prescribed for the management of pain and inflammation, but their conventional oral administration is often associated with gastrointestinal irritation, ulceration, and systemic adverse effects. These limitations have encouraged the development of alternative drug delivery approaches that can provide localized action while minimizing systemic exposure. Topical drug delivery systems have emerged as a promising strategy, offering direct application at the site of inflammation and improved patient compliance. However, conventional topical formulations such as creams and gels frequently suffer from poor drug penetration and short duration of action.
Microsponge-based drug delivery systems represent an advanced approach designed to overcome these challenges. These systems consist of porous polymeric particles capable of entrapping active pharmaceutical ingredients and releasing them in a controlled manner over an extended period. When incorporated into a topical gel, microsponges can enhance drug stability, prolong release, and reduce local irritation. Diflunisal, a salicylate NSAID with potent anti-inflammatory activity and a relatively long half-life, is considered a suitable candidate for such delivery systems.
This review focuses on the principles of microsponge technology, its application in topical drug delivery, and its potential to improve the therapeutic performance of Diflunisal by ensuring sustained release and targeted action.
