Abstract :
The Alzheimer’s disease (AD) is a neurodegenerative disease and leading cause of dementia which involves progressive decline of cognitive function and behavioral disorders. The rising life expectancy and increase in elderly population have greatly increased the burden of AD on public health. Currently available treatment modalities, like cholinesterase inhibitors, N-methyl-D-aspartate (NMDA) receptor antagonists and anti-amyloid monoclonal antibodies offer relief from symptoms or delay in disease progression but lack efficacy in prevention of neurodegeneration. With the advancement of knowledge about the molecular mechanism of disease process, a large number of therapeutic targets have been identified, namely amyloid beta, tau protein, neuroinflammation, oxidative stress, mitochondrial dysfunction, synapse loss, and insulin resistance. Modern treatment strategies include gene therapy, RNA based therapeutics, stem cell therapy, immunotherapy, personalized medicine, repurposed drugs and novel drug delivery systems. More research and clinical trials will lead to development of safer and efficacious therapies to manage the disease process and alleviate the burden of the disease.
