Abstract :

Alzheimer’s disease is a progressive neurodegenerative disorder associated with cognitive decline, memory impairment and neuronal dysfunction. Oxidative stress is considered an important factor associated with neuronal damage and cognitive impairment. The present study was conducted to evaluate the antioxidant and neuroprotective potential of Thespesia populnea fruit extract. The fruit was extracted with methanol using Soxhlet extraction and subjected to preliminary phytochemical screening. The in-vitro antioxidant activity was evaluated using DPPH, phosphomolybdenum and ferric reducing assays. Neuroprotective activity was evaluated against aluminium chloride-induced cognitive impairment in Wistar albino rats. The extract was administered at 200 and 400 mg/kg and behavioural changes were assessed using Elevated Plus Maze, Cook’s Pole Test and Y-Maze. Catalase activity, total brain protein and histopathological changes were also evaluated. The extract showed concentration-dependent antioxidant activity and improved learning and memory in treated animals. The treatment also improved biochemical parameters and reduced neuronal alterations in brain tissue. Overall, the findings suggest that Thespesia populnea fruit extract possesses antioxidant and neuroprotective potential against aluminium chloride-induced cognitive impairment.
