Abstract :

Coumarin and its derivatives are a prominent class of benzopyrone compounds known for diverse pharmacological properties, including anticancer, antimicrobial, and antioxidant activities. The present study focuses on the synthesis, characterization, and evaluation of anticancer activity of novel coumarin derivatives. The synthesis was carried out through Pechmann condensation and subsequent functional modifications at various positions of the coumarin nucleus to enhance biological efficacy. The synthesized compounds were characterized by FT-IR, ¹H-NMR, ¹³C-NMR, and Mass spectroscopic techniques to confirm their structural integrity and purity. The anticancer potential of the synthesized derivatives was assessed using MTT assay against selected human cancer cell lines, such as MCF-7 (breast cancer), HeLa (cervical cancer), and A549 (lung cancer). Results revealed that several derivatives exhibited significant cytotoxicity, which may be attributed to the presence of electron-donating or withdrawing substituents influencing the coumarin core. The findings suggest that structural modification of coumarin scaffolds can lead to promising lead molecules for future anticancer drug development.
Keywords: Coumarin derivatives, synthesis, characterization, anticancer activity, MTT assay, structure–activity relationship.
