Abstract :

Hyperpigmentation is a persistent and multifactorial skin disorder characterized by excessive melanin deposition and associated with melasma, post-inflammatory hyperpigmentation, solar lentigines, freckles, and photoaging. Conventional depigmenting agents and procedures are often limited by poor skin penetration, instability, irritation, recurrence, and inconsistent efficacy. This review evaluates the potential of nanotechnology, focusing on nano emulsions, metallic nanoparticles, and hybrid nano emulsion metallic nanoparticle systems for improved management of hyperpigmentation. The molecular mechanisms of pigmentation are discussed, with emphasis on tyrosinase, microphthalmia-associated transcription factor, tyrosinase-related protein 1, tyrosinase-related protein 2, melanocortin 1 receptor signaling, cyclic adenosine monophosphate signaling, mitogen-activated protein kinase signaling, and Wingless-related integration site/β-catenin signaling, together with the roles of oxidative stress, inflammation, melanosome biogenesis, and melanosome transfer. Nano emulsions can enhance solubilization, stability, skin permeation, bioavailability, and controlled release, whereas metallic nanoparticles provide antioxidant, anti-inflammatory, ultraviolet-protective, and melanogenesis-modulating effects. Their integration may improve dispersion, delivery, retention, stability, and targeting while reducing irritation. Future directions include artificial intelligence-assisted formulation, personalized nano therapy, exosome-mediated delivery, stimuli-responsive systems, advanced skin models, green nanotechnology, and clinical translation. However, further research is required to establish long-term safety, optimize particle characteristics, validate efficacy through clinical studies, and facilitate reproducible manufacturing, regulatory approval, and successful commercialization for broader therapeutic and cosmetic applications.
