Abstract :

The present research work focuses on the formulation and development of extended-release tablets of Cyproheptadine Hydrochloride using natural polymers with the objective of achieving prolonged drug release, reducing dosing frequency, and enhancing patient compliance. Natural polymers such as guar gum, xanthan gum, and gum karaya were selected as release-retarding agents due to their biocompatibility, cost-effectiveness, and safety.
Pre-formulation studies, including drug-excipient compatibility tests, were carried out to ensure stability of the formulation. Extended-release tablets were prepared by the direct compression/wet granulation method and evaluated for various pre-compression parameters (bulk density, tapped density, angle of repose) and post-compression parameters (hardness, friability, weight variation, and drug content), all of which were found within pharmacopeial limits.
In-vitro dissolution studies revealed that the drug release rate varied with the type and concentration of natural polymers used. Among the formulations, the optimized batch exhibited a sustained drug release up to 12 hours, following near first-order release kinetics with a non-Fickian diffusion mechanism.
The study concludes that natural polymers can be effectively utilized in the design of extended-release matrix tablets of Cyproheptadine Hydrochloride, providing a promising alternative to synthetic polymers for achieving controlled drug delivery, better therapeutic efficacy, and improved patient compliance.
