Abstract :

The present study aimed to Formulate and Evaluate mouth dissolving tablets (MDTs) of Perampanel to enhance patient compliance and achieve rapid onset of action. Mouth dissolving tablets were prepared by the direct compression method using super disintegrants such as Crospovidone and Croscarmellose sodium, along with Avicel PH 102 as a directly compressible diluent. Various formulations were developed and evaluated for both pre-compression and post-compression parameters.
Pre-compression studies, including angle of repose, bulk density, tapped density, Carr’s index, and Hausner’s ratio, indicated satisfactory flow properties of the powder blends. Post-compression evaluation of tablets, such as weight variation, hardness, thickness, friability, drug content uniformity, wetting time, disintegration time, and in vitro dissolution studies, demonstrated that all formulations complied with Indian Pharmacopoeial (IP) limits.
Among the developed formulations, F8 exhibited superior performance with rapid disintegration and maximum drug release of 99.95% within 30 minutes, and was therefore considered the optimized formulation. The enhanced dissolution was attributed to the synergistic effect of Crospovidone and Croscarmellose sodium, which promoted rapid water uptake and tablet disintegration, while Avicel PH 102 provided adequate mechanical strength and compressibility.
In conclusion, the study successfully developed Perampanel mouth dissolving tablets with desirable physicochemical properties and rapid drug release, indicating their potential as an effective alternative dosage form for improving patient compliance.
