DESIGN AND DEVELOPMENT OF ORAL FLOATABLE IN-SITU GEL FOR INVASIVE FUNGAL INFECTION

Abstract :

In the present study in-situ oral floatable gelling system of Posaconazole was successfully formulated by using different concentrations of gellan gum and pectin. Total eleven formulations were prepared with variable concentration of gellan gum and pectin After performing the evaluation tests of prepared formulations, it can be summarized that Batch F9 was selected as best formulation, because it shows good release property of drug from the gel, having sufficient gel strength, shows optimum pH required for sol-gel conversion, and it also exhibit the acceptable result of other parameters. The release pattern of the optimized formulation (F9) was best fitted to a first order. The mechanism of the drug release was non-Fickian (super case II) transport. Non-Fickian (super case II) transport mechanism for releasing a drug associated with stresses and state-transition in hydrophilic glassy polymers, which swell in water or biological fluids. This term also includes polymer disentanglement and erosion. In-situ forming polymeric formulations drug delivery system is in a sol form before administration in the body, but once administered, it undergoes gelation in-situ to form a gel. The findings showed that the oral floatable in-situ gel offers a better sustained property compared to a conventional dosage form.
