FORMULATION AND EVALUATION OF SELF MICROEMULSIFYING DRUG DELIVERY SYSTEM OF ROSUVASTATIN CALCIUM DRUG

Abstract :

Rosuvastatin Calcium is an anti- hyperlipidemic drug. It is mainly employed to treat hypercholesterolemia. According to the Biopharmaceutical Classification System (BCS), this medication is classified as a class II medicine because of its crystalline form, which results in low dissolving and a 20% oral bioavailability. This study’s primary goal is to create a self-microemulsifying drug delivery system (SMEDDS) for calcium rosuvastatin to increase its solubility. In this study, liquid SMEDDS (L-SMEDDS) containing rosuvastatin along with sesame oil and Olive oil was formulated. Rosuvastatin Calcium solubility analysis was used to generate ternary phase diagrams in order to optimize the system. The oil phase consisted of a 4:1 mixture of sesame oil and olive oil, whereas the surfactant and co-surfactant were PEG 400 and Tween 20 respectively. The prepared L-SMEDDS formulation were evaluated for, globule size and zeta potential, % transmittance, FTIR study, drug content and in-vitro drug release profile.
According to the results, the L-SMEDDS formulation has a higher drug content and a higher rate of drug release in less time than the Rosuvastatin API. L-SMEDDS’s reconstitution properties revealed no evidence of phase separation and spontaneous self-micro emulsification. Globule size analysis F1 showed globule size 205nm and zeta potential -7.8 mV. The in-vitro drug release results demonstrated a significant improvement in rosuvastatin’s rate of dissolution. The F1 formulation’s transmittance percentage at all times is 98%, indicating good surfactant and co-surfactant emulsification.
Since the self-micro emulsified formulation was more soluble than the API L-SMEDDS, it seemed like a promising solution to the issues surrounding rosuvastatin oral administration.
