Abstract :
The development of advanced drug delivery systems has revolutionized pharmaceutical science, with oral dispersible films (ODFs) emerging as a promising alternative to traditional solid dosage forms. ODFs are thin, flexible polymeric strips that disintegrate rapidly in the oral cavity without the need for water, offering a patient-friendly solution particularly suited for pediatric, geriatric, and dysphagic populations. This thesis explores the formulation, characterization, and potential of ODFs as a modern drug delivery platform.
Composed of film-forming polymers, plasticizers, active pharmaceutical ingredients, and excipients like sweeteners and flavoring agents, ODFs are designed for rapid disintegration, improved palatability, and effective drug delivery. The oral cavity, particularly the buccal and sublingual mucosa, provides an ideal absorption site due to its vascularity and ability to bypass first-pass metabolism, thus enhancing bioavailability.
The thesis also reviews the historical evolution of ODFs, tracing their development from early orally disintegrating tablets to current innovations involving nanotechnology and mucoadhesive systems. Advantages such as rapid onset of action, enhanced patient compliance, and precise dosing are weighed against formulation challenges like drug-excipient compatibility, moisture sensitivity, and scalability. In conclusion, ODFs represent a significant advancement in oral drug delivery, combining convenience, efficacy, and innovation. With continued research and technological integration, they hold the potential to transform therapeutic outcomes and expand treatment accessibility, particularly for vulnerable patient groups. This thesis aims to contribute to the growing body of knowledge surrounding ODFs and support their adoption in mainstream pharmaceutical practice.
