NEXT-GENERATION EXCIPIENTS: A COMPREHENSIVE REVIEW OF INNOVATION IN DRUG DEVELOPMENT

Abstract :

Novel excipients are quickly important to overcome formulation challenges in advanced drug distribution systems. Despite regulatory obstacles and unclear definitions, their role is expanded in improving stability, solubility and bioavailability. Pharmaceutical excipients are involved in a drug formulation that is not entitled to medical activity, but plays an important role in ensuring effective distribution, stability and production of active pharmaceutical ingredient (API). As defined by Pharmacopeia in the United States, the materials that are deliberately added to drug products and have been evaluated for security.Drug products not only contain “actives” that confer the intended therapeutic benefits such as pain relief or act on particular part of the body, but contain other materials that are also “functional” with respect to the drug product. These are known as excipients and specific functionality which they confer to a particular product is independent upon the process used to add the excipient to the formulation and its exact location within the final dosage form. Introduction of novel drug delivery systems and new drug. moieties lead to the need for new excipients with varied characteristics. Development of new excipient entities and their evaluation is a costly procedure; modification. The field of pharmaceutical formulation is constantly evolving, driven by the need for improved drug delivery systems and enhanced therapeutic outcomes. One critical aspect of this evolution is the development and utilization of novel excipients. This review explores the types and examples of novel excipients that have revolutionized drug formulation in recent years. These compounds, in addition to their basic function as excipients, may have additional properties, e.g. Stimuli sensitivity,enzyme inhibition, intestinal epithelium penetration enhancement, efflux pump inhibition, taste-masking, pharmacological activity and the ability to interact with enzymes responsible for drug metabolism.
